Culture methods

Culture Media based on Solid or Liquid

  1. Solid medium - agar 1.5 -2
  2. Semi-solid - agar <0.5 for motility
  3. liquid medium = broth

Methods of Applying Organisms to Medium

  1. Streak Culture(streak a bacteria) - Isolation from mixed population
  2. lawn/Carpet culture(like garden full of bacteria,then add antibiotic to find suspectibilty)- Anti-biotic sensitivity
  3. stab culture/puncture - O2 requirement,motility(crag's tube method)

  4. Anaerobic Culture Method

    1. Candle Jar(old)
    2. Macintosh Field anaerobic jar
    3. Gas pak system(simple method chemical observes Oxygen from Air )

Based on Composition of Nutrients

  1. Basal medium(white rice) = BAsic nutrients no special nutrients

  2. Complex Medium (multiple nutrients) eg:(brain heart infusion agar )

  3. Defined Medium(exact amount of nutrients) = eg; ALkalline peptone water

BAsed on Nutrients for Particular Bacteria

  1. Special Medium
    • Enriched Medium(enriched with spl items) , if known composition Defined medium , if unknown Complex medium
    • Enrichment Medium only in broth form (Only als growth of particular organisms)
    • Selective Medium only Solid form(help grown one Organism than other)
    • Differential medium(help differentiate organism) eg: urease medium , mc-conkey agar
    • Indicator medium (Indicates reaction) eg;Mc-Conkey color change
    • Transport Medium/Holding Medium (stop multiplying, stop dying) eg: cary-blair medium and VR medium for Cholera ,Pikes medium -strep.pyogenes,Stuart and Ame's for Gonococcal
    • Fermentation medium(Distinguish fermentation type)
    • For Anaerobes
      1. Robertson Cooked Meat medium
      2. PRAS - PRe reduced Anaerobicaly Sterilised Medium
